In addition to what is mentioned above, I remember reading that the air cooling of the engine contributes to the sound as well. Possibly a water cooled engine buffers the sound a bit.

the distinctive harley sound is really due to the single crankshaft pin.... from the link above:
The difference in the Harley engine is that the crankshaft has only one pin, and both pistons connect to it. This design, combined with the V arrangement of the cylinders, means that the pistons cannot fire at even intervals. Instead of one piston firing every 360 degrees, a Harley engine goes like this: * A piston fires. * The next piston fires at 315 degrees. * There is a 405-degree gap. * A piston fires. * The next piston fires at 315 degrees. * There is a 405-degree gap. And the cycle continues. At idle, you can hear the pop-pop sound followed by a pause. So its sound is pop-pop...pop-pop...pop-pop. That is the unique sound you hear!

Sometimes it's nitromethane that gives those 2 big cylinders that deep rumble.

Nobody has mentioned stroke. A Harley has a longer stroke compared to most other engines. This is why the ricers are so slow off the line. Their stroke is shorter which keeps them from making power in the low rpm range. It is a battle between torque and horsepower with HD being a torque base engine and the shorter stroke engines being HP based engines. There is always a trade off between the two. The potato, potato sound is the added bonus.

It does it because of cylinder displacement. If you took my 96" V-2 and multiplied it by four you would get a 384" V-8 and it would rumble. If you took a six cylinder Camaro and cut off 4 cylinders you would only have a 48 CI V-twin and that would not sound like a Harley, it would sound like a Honda copy of a Harley because it would only have half of the displacement per cylinder. 3 Harley cylinders equal 1 Camaro V-6 displacement wise, but each cylinder in the V-8 is almost the same as the Harley.

V6= 3600cc/16.393=219.6CI/6=36.6CI per cylinder x2=73.2CI. Should sound like a 1200. Maybe need facial hair and tats?
