Skip shift and a tune?

Can You override skip shift with a tune or do you have to purchase an eliminator to physically jump the circuit on the 2010 M6 transmission?

Yes a tune can disable skip shift.

Just wait until after 2500 rpms before shifting to 2nd. That's the Free Ship Shift!
