That's probably not a good idea. I don't know this for sure, but I imagine that, unless you're driving on always smooth as glass roads, the bumps won't be good for the disk and you'll end up losing information and having to buy another hard drive. Better to stick with flash memory.

Not saying I have 500GB of music, just that my music is on a 500GB hard drive (although if recorded in a "lossless" format, they can take up good amount of room). I didn't think about the vibration, as that would be an issue unless it as a SSD.
I guess the better question then is what file system(s) does it support? FAT, FAT32, NTFS, HFS, HFS+... ?

Also, the smaller ipods (<16Gb, I believe) are flash memory, so no moving parts. A big external harddrive is going to have platters that could get corrupted from excessive vibration. SSD would definitely be ok though.

You could just mount the drive vertically to reduce vibrations.
